چکیده
objective: the purpose of this study was to investigate the role of fast inactivating (ia) voltage dependent potassium channels in shaping the discharge activity of cerebellar purkinje cells (pcs) in ataxic rats. materials and methods: male sprauge-dawley rats (40-60 g) were used in this study. ataxia was induced via single i.p. injection of 3-acethylpyridine (65 mg/kg). four days following induction of ataxia, the rats were decapitated after being anesthetized using ether. 300 μm thick parasagittal slices were prepared from the vermis of the cerebellum. sharp intracellular recording was achieved from purkinje cells under continuous superfusion of acsf containing synaptic blockers. results: intracellular recording of 21 purkinje neurons in cerebellar slice preparation showed that purkinje cells of ataxic rats fire in three modes of tonic (23.81%), burst (19.05%) and silence (57.14%). following the application of 4-ap, 83.33% of the silent pcs were spontaneously fired and the burst activity was robustly enhanced in bursting pcs. in addition, 4-ap significantly increased the duration of burst and active periods but decreased the duration of inactive period. it also increased the number of sodium spikes within each burst. conclusion: 4-ap sensitive current in purkinje cell of ataxic rat involves in three modes of firing pattern. in addition, the inhibition of fast inactivating potassium channels (ia) with 4-ap enhances the spontaneous discharge activity of purkinje cells.

Elevated Neuronal Excitability Due to Modulation of the Voltage-Gated Sodium Channel Nav1.6 by Aβ1−42
Aberrant increases in neuronal network excitability may contribute to the cognitive deficits in Alzheimer's disease (AD). However, the mechanisms underlying hyperexcitability are not fully understood. Such overexcitation of neuronal networks has been detected in the brains of APP/PS1 mice. In the present study, using current-clamp recording techniques, we observed that 12 days in vitro (DIV) pr...
متن کاملThe sodium channel accessory subunit Navβ1 regulates neuronal excitability through modulation of repolarizing voltage-gated K⁺ channels.
The channel pore-forming α subunit Kv4.2 is a major constituent of A-type (I(A)) potassium currents and a key regulator of neuronal membrane excitability. Multiple mechanisms regulate the properties, subcellular targeting, and cell-surface expression of Kv4.2-encoded channels. In the present study, shotgun proteomic analyses of immunoprecipitated mouse brain Kv4.2 channel complexes unexpectedly...

Many neurotransmitters regulate action potential activity in neuronal, endocrine, and cardiac cells by rapidly modulating the gating of K+ channels. Neurotransmitters might also produce prolonged effects on excitability by regulating the expression of K+ channel genes. Here we show that the neuropeptide thyrotropin-releasing hormone (TRH) downregulates Kv1.5 and Kv2.1 K+ channel mRNAs in clonal...
متن کاملCell adhesion molecule L1 contributes to neuronal excitability regulating the function of voltage-gated Na+ channels.
L1 (also known as L1CAM) is a trans-membrane glycoprotein mediating neuron-neuron adhesion through homophilic and heterophilic interactions. Although experimental evidence has implicated L1 in axonal outgrowth, fasciculation and pathfinding, its contribution to voltage-gated Na(+) channel function and membrane excitability has remained unknown. Here, we show that firing rate, single cell spikin...
